About collection: The collection includes 6 specialized collections keeping isolates of viruses and strains of bacteria, fungi and microorganisms similar to fungi important for agriculture for different reasons. Workers of Collection take part important role in the Czech “The National Programme on Conservation and Utilization of Microbial Genetic Resources Important for Agriculture“ based by Ministry of Agriculture of the Czech Republic. The specialized collections were arising mostly during last 30 years in different departments of Crop Research Institute following of solved research activities.

Number of strains: At present time, the collection holds approximately 90 isolates of viruses, more than 800 strains of bacteria, more than 2000 strains of fungi and microorganisms similar to fungi (Oomycota). The vast majority of them are hold within Czech National programme on conservation and utilization of microbial genetic resources important for agriculture.

The different methods of conservation are used for storage of the microorganisms in collection. Different groups of microorganisms are conserved by different types of preservation. The strains are preserved in liquid nitrogen, and in deep freezer at temperature-80 °C, in form freeze-dried conserves, under paraffin oil on an agar slant, in explantate plants and on live plants in glasshouse.

Characterization of collection: The collection preserves strains of leaf rust (Puccinia triticina), stem rust (Puccinia graminis f. sp. tritici), yellow rust (Puccinia striiformis f. sp. tritici), crown rust (Puccinia coronata), powdery mildew (Blumeria graminis f.sp. tritici) and common bunt (Tilletia laevis, Tilletia caries) and dwarf bunt (Tilletia controversa). The collection items are used to test resistance of varieties and new wheat breeding lines in the greenhouse and field conditions. Strains are used to determine the genetic basis of resistance and for breeding purposes, passportization of wheat and oat varieties and for research purposes. Isolates of phytopathogenic microorganisms are also used in international cooperation in epidemiological studies and determination of pathogen variability.

Number of strains: At present, the collection contains more than 1 000 strains of rusts and powdery mildew. The collection actively participates in the “National programme on conservation and utilization of genetic resources of microorganisms and small animals with economic importance”.

Methods of culture maintenance: The strains are stored by cryopreservation in liquid nitrogen and in ultra-low temperatures (-85 °C), strains of powdery mildew are stored on live plants. Strains of bunt are stored in ultra-low temperature (-85 °C) and in a refrigerator at 5 – 8°C.

Services: The collection offers identification of rusts and its pathotypes, inoculum propagation for infectious experiments, advice on taxonomy and crop protection, and long-term storage of urediospores.

Characterization of collection: The Collection includes a group of quarantine, phytopathogenic and potentially pathogenic bacteria that cause economic losses in crops, vegetables, fruit, ornamental and forest trees and ornamental plants in the Czech Republic. The second part of the Collection includes agriculturally beneficial bacteria isolated from phyloplane of a wide range of plants in the Czech Republic with antagonistic properties against pathogens, with anti-stress and stimulating properties supporting health, growth and sustainable crop production. The characteristics of all deposited strains are available on the website of the National Program.

Number of strains: At present, the collection approximately 270 strains of bacteria. The strains are maintained under the Czech “The National Programme on Conservation and Utilization of Microbial Genetic Resources Important for Agriculture“.

Methods of culture maintenance: All strains are stored frozen at -80 to -90°C and as freeze-dried cultures at 4°C. Strains that are biologically hazardous substances are also stored in liquid nitrogen.

3. Collection of plant pathogenic viruses

Characterization of collection: In the collection, there are maintained economically important viruses of cereals, vegetables, fruit trees, hop and grapevine. Individual isolates of the collection are used by an expert diagnosis as comparative standards. They are used for research work e.g. for testing resistant plants and offered to other institutions. The isolates are gradually characterized by the use of molecular biological methods. The collection take part in Czech “The National Programme on Conservation and Utilization of Microbial Genetic Resources Important for Agriculture“.

Number of strains: The collection contains 89 isolates of plant viruses and three strains of bacteria causing European stone fruit yellows phytoplasma (ESFY). The total number of the collection items changes as new isolates are collected from the nature.

Methods of culture maintenance: Plant viruses are stored dried in host plant leaves at 2-6 °C or they are preserved frozen in leaves at -70 °C, by freeze-drying and freezing in liquid nitrogen. Viruses which lose their infectiousness by drying or freezing are maintained and regularly inoculated to new indicator plants mechanically or by the use of insects like aphids (Myzus persicae, Rhopalosiphum padi) or a leafhopper (Psammotettix alienus). Infected plants are maintained in isolated greenhouse compartments or on woody plants in a screen house.

Services: The isolates of the collection are used as positive standards by diagnosis of cereal, vegetable, fruit tree and grapevine viruses. They serve as a source of infectious material for testing possibly resistant plants, evaluating the immunity against plant viruses and for diagnosis by the comparison of symptoms on indicator plants. The viral isolates are used for solving of a research work. Samples of the isolates of the collection are available to other research and diagnostic institutions of the Czech Republic and other countries when the regulations of Material Transfer Agreement are observed.

4. Collection of edible and medicinal macromycetes

Characterization of collection: The collection preserves in vitro cultures of edible and medicinal macromycetes from the groups Ascomycota and Basidiomycota, which are of importance for potential production cultivations. The most numerous part of the collection represent morel fungi (especially genera Morchella and Verpa), the other part is represented by various species of edible and / or medicinal mushrooms from the group Basidiomycota. The collection actively participates in Czech “The National Programme on Conservation and Utilization of Microbial Genetic Resources Important for Agriculture“.

Number of strains: At present, approximately 140 strains are included in the collection. The strains are maintained under the Czech “The National Programme on Conservation and Utilization of Microbial Genetic Resources Important for Agriculture“.

Methods of culture maintenance: The strains are preserved in liquid nitrogen, on an agar slant under paraffin oil and on a rye substrate.

List of fungal strains

Services: The collection provides fungal strains to potential users for the research purposes. The collection also provides consultations on the maintenance, storage and cultivation of edible and medicinal macromycetes (e.g. at mycological exhibitions, etc.). Selected strains are used to demonstrate the cultivation of edible and medicinal macromycetes during excursions of university and high school students.

5. Collection of Soil Bacteria

Characterization of collection: The Collection of soil bacteria originated from the Collection of Microbe Cultures, which was founded by prof. V. Káš in 1948. It was later renamed on the Collection of Rhizobia, which forms the basis of the Soil Bacteria Collection. At present, the collection preserves in vitro cultivable soil bacteria of the genera Rhizobium, Bradyrhizobium, Sinorhizobium and Azotobacter. Isolates of these bacteria were obtained from the scientific work of the Agricultural Soil Science and Pedobiology and from other institutes and collections. Cultures are preserved in agar tubes and at the same time in the form of freeze-dried conserves. Strains stored in the Collection are a source for the work of various scientific institutes and universities. The collection actively participates in the National Program for the Conservation and Utilization of Genetic Resources of Microorganisms and Small Animals of Economic Importance.

Number of strains: The collection contains approximately 550 strains of bacteria, which are stored under the Czech “The National Programme on Conservation and Utilization of Microbial Genetic Resources Important for Agriculture“.

Methods of culture maintenance: Strains are preserved in form of freeze-dried conserves and on an agar slant in test tubes.

Characterization of collection:

The collection holds strains of fungi from groups Ascomycota, Basidiomycota, Zygomycota and microorganisms from group Oomycota cultivable in in vitro conditions. The fungi are pathogens or potential pathogens of plant or edible or medicinally mushrooms. The collections keeps mycotoxinogenic or potential mycotoxinogenic fungi, fungi useful or potentially useful in plant protection or for detection of harmfully microorganisms of plant, fungi damaging stored agriculturally products or edible and medicinal mushrooms . Collection takes active part in Czech “The National Programme on Conservation and Utilization of Microbial Genetic Resources Important for Agriculture“.

Number of strains: At present time, the collection holds more than 710 strains of fungi or microorganisms similar to fungi (Oomycota). More than 630 of them are hold within Czech “The National Programme on Conservation and Utilization of Microbial Genetic Resources Important for Agriculture“.

Methods of culture maintenance: The strains are preserved in liquid nitrogen, in form freeze-dried conserves (only the strains creating spores and yeasts) and under paraffin oil on an agar slant in test tubes.
